For example if I have 4 martini glasses can I use calculated shipping and if a customer buys one, it is the amount in the shipping calculator. If they buy 3 can I add a rule that will charge $5 for each additional? Or even better a percentage?
If so, is it possible to then charge a different amount if the items are small and will only cost an additional $2.00 per item?
At the moment I am only selling in Canada and have Canada Post and UPS as options.

Yes the calculations for combined shipping happen on the cart. eBay is a bit unusual, most sites don't show ANY shipping information until you go to checkout.

If you use calculated shipping eBay will automatically calculate the charge for each additional item from the same listing.
Be aware there is a flaw in the system, eBay will double the weight used to calculate but they do not increase the dimensions. If the additional items can all fit in the same box as a single it's not a problem, if the box needs to be bigger but not so big as to trigger dimensional weight calculations it's not a problem. If the multiple items require a bigger box that does trigger dimensional weight then the calculation may not be correct (too low).

Thank you recped for your answer, I appreciate the detailed reply. I have done that in the past but it doesn't always work the best depending on how many items I have available, and their size. From the eBay listings I have been checking it seems to be only available when using flat rate - adding $5 (for example) for each additonal item - and because my shipments are usually in large boxes it can make a big dollar difference to the customer from East to West coast by using calculated.
I am actually using it on a few listings like Park Ridge by Mikasa for 2 sets of 4 glasses but one issue I have is that I cannot find out how much a customer would be quoted for shipping if they want both sets. Is there some way that I can see what someone in say Toronto would pay for shipping if they want both sets?

Just look at your listing (as a buyer) but change your ship to Postal Code to one from Toronto (or any other location)

Thankyou recped, I do appreciate your time. Unfortunately that does not seem to work. When I try to see what two sets would cost, the shipping estimate remains the same. The pop up does not show it is for 2 items and there does not seem to be a way to update it from one to two for an estimate. I asked a friend to check and their findings were the same.
My settings are box size 28 x 28 x 28 cm at 2.5 kg which is 3.66 kg volumetric.
Two sets should trigger it to 5 kg which puts the volumetric box size at 31 x 31 x 31.
The difference is not as much as I thought, though. Using my small business account and Canada Post Expedited it is only a difference of about $6 to Ontario and of course with eBay's discounts it would be less but that is not really the point.
I found another listing similar to mine in the U.S. - it has 10 items available - and it also does not change the estimate no matter how many I enter in to the little box. I realize that this is only the estimate and once a customer adds to cart or clicks buy it now, the information would probably be available (and of course I can't check my own listing by doing that).
The estimate used to be available quite easily if memory serves (but that is a whole other conversation lol).

I just checked the U.S. listing again and it does say add to cart to see what the combined shipping discount would be, so that may just be the case. No estimate without adding to cart.
